Taking amitriptyline and Valdoxan (agomelatine) together can increase the levels of amitriptyline in your blood. This may lead to an increased risk of side effects such as extreme drowsiness, dry mouth, blurred vision, constipation, or a fast heartbeat. There is also a small risk of a rare but serious condition called serotonin syndrome, which can cause shivering, sweating, and confusion.

Why this happens
Valdoxan can interfere with the way the liver breaks down amitriptyline (specifically through an enzyme called CYP1A2). This causes amitriptyline to build up in the body. Additionally, both medicines affect brain chemicals, which can lead to additive side effects.
What you should do
